Size-exclusion chromatography

The size-exclusion chromatography column is filled with the sieving medium, which enables separation of the sample components according to their hydrodynamic diameter. During the gravity flow, larger particles, such as extracellular vesicles, are eluted earlier, while smaller molecules, including salts and polysaccharides, are retained longer within the column matrix.

This mild, aqueous-based separation technique preserves the structural and functional integrity of vesicles. Size-exclusion chromatography was found to be efficient in extracellular vesicle isolation from biological samples of various origins and is now one of the most frequently used techniques for the isolation of extracellular vesicles due to its selectivity, scalability, and reproducibility.
